{"id":222,"date":"2014-03-21T02:53:13","date_gmt":"2014-03-21T02:53:13","guid":{"rendered":"http:\/\/joshmitteldorf.peachpuff-wolverine-566518.hostingersite.com\/?p=222"},"modified":"2014-03-25T19:23:03","modified_gmt":"2014-03-25T19:23:03","slug":"a-one-man-experiment-in-radical-anti-aging","status":"publish","type":"post","link":"https:\/\/scienceblog.com\/joshmitteldorf\/2014\/03\/21\/a-one-man-experiment-in-radical-anti-aging\/","title":{"rendered":"A one-man experiment in radical anti-aging"},"content":{"rendered":"<p dir=\"ltr\"><em>One reason that there is still so much uncertainty in anti-aging medicine is that we can\u2019t do experiments on humans the way we do on mice. \u00a0So we are grateful for a few human guinea pigs who put their own bodies on the line as early adopters. \u00a0There is the <a href=\"https:\/\/www.crsociety.org\/\">Caloric Restriction Society<\/a>, people who are losing weight in the hope of gaining years. \u00a0There is an on-line community of people experimenting with <a href=\"http:\/\/c60.net\">Buckyballs in olive oil<\/a> (this may sound like a joke if you haven\u2019t read about it before).<\/em><\/p>\n<p dir=\"ltr\"><em>And then there is Jim Green of Wichita, KS. \u00a0Since 2007, Jim has been pursuing experimental strategies and claims to have set his body clock back by 15 to 20 years already. \u00a0The core of his program is telomerase activation herbs, particularly astragalus, taken in much larger quantities than the label recommends. \u00a0Jim has quite a story to tell, and this week I have interviewed him, an exclusive for ScienceBlog.<\/em><\/p>\n<p dir=\"ltr\">(You can read more about Jim&#8217;s career <a href=\"http:\/\/www.greenray4ever.com\/index-88.html\" target=\"_blank\">here<\/a> and his anti-aging program <a href=\"http:\/\/www.greenray4ever.com\/longevity.html\" target=\"_blank\">here<\/a>.)<\/p>\n<figure style=\"width: 360px\" class=\"wp-caption alignleft\"><img loading=\"lazy\" decoding=\"async\" class=\"  \" alt=\"\" src=\"https:\/\/lh6.googleusercontent.com\/8_RsfUDyGVsjDZc4zGGmx-4zq3blq-Ugrfa8rbQoB-re-0NDlVVb0o6muvy-jUKClgGwsb5JHlALMiBmsEEFBj0A8d3FOLYQShykjJtKWZDUXSNC1BdRvFVUPBUk\" width=\"370\" height=\"490\" \/><figcaption class=\"wp-caption-text\">Jim Green in 2007<\/figcaption><\/figure>\n<figure style=\"width: 268px\" class=\"wp-caption alignright\"><img loading=\"lazy\" decoding=\"async\" alt=\"\" src=\"https:\/\/lh4.googleusercontent.com\/LUqHJzY9WuX6XuXbsTVO9jKPEGT-Ia1jmPK4LL8ewggnnrb6nTerxw79I2te0aJK0K_bttAHlOwRcTSF0TFi2EvQ7QAh_EY7WZRMsb6--ML0Vk_1bq_LKToibta3\" width=\"278\" height=\"344\" \/><figcaption class=\"wp-caption-text\">Jim Green in 2013<\/figcaption><\/figure>\n<p dir=\"ltr\"><strong>What gave you the idea that growing younger was possible?<\/strong><\/p>\n<p dir=\"ltr\">According to the\u00a0<a href=\"http:\/\/www.greenray4ever.com\/longevity2.html#TELOMEREPOSITIONINGEFFECT\">telomere position effect <\/a>data I saw early on back in 2007, telomere length impacts youthful gene expression, and gene expression is more youthful the longer the telomeres are. Furthermore, since 1998 it has been realized that<a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4cb.html#CELLULARSENESCENCE\"><span style=\"color: #000000\"> senescent cells<\/span><\/a> can often be restored to the youthful phenotype by transfecting them with a virus that improves hTERT transcription and boosts levels of<a href=\"http:\/\/www.greenray4ever.com\/longevity2.html#(7)\"> telomerase<\/a>e in the cell. In 1998, <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4f.html#FOSSELMICHAEL\">Michael Fossel <\/a>published <a href=\"http:\/\/online.liebertpub.com\/doi\/pdfplus\/10.1089\/rej.1.1998.1.3\">an article<\/a> on this in the Journal of Anti-Aging Medicine (now Rejuvenation Research). [<a href=\"http:\/\/jama.jamanetwork.com\/article.aspx?articleid=187604\">Another Fossel article<\/a> -JJM] So there were definite grounds for optimism that lengthening telomeres with telomerase could result in rejuvenation transformations.<\/p>\n<p dir=\"ltr\">On the other hand, as telomeres got shorter, patterns of gene expression became more elderly and the likelihood of the onset of<a href=\"http:\/\/www.greenray4ever.com\/lifexfoot2.html#103b\"> diseases of old age<\/a> increased, according to many scientific studies. For instance, microglial cells in the brain that clean up <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4am.html#AMYLOIDBETA\">amyloid beta <\/a>plaque leading to<a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4am.html#ALZHEIMERSDISEASE\">Alzheimer&#8217;s diseas<\/a>e fail when they become <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4r.html#REPLICATIVESENESCENCE\">senescent<\/a>. If senescence in the microglial cells (derived from hematopoietic stem cells) could be prevented, then Alzheimer&#8217;s disease due to the onset of senility might also be prevented. Similarly, when the lining of the vascular endothelium becomes senescent, it becomes more adhesive to monocytes and more likely to develop<a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4an.html#ATHEROSCLEROTICPLAQUE\"> atherosclerotic plaque<\/a> leading to<a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4hs.html#HEARTATTACK\"> attack<\/a>s o<a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4sm.html#STROKE\">r stroke<\/a>s. \u00a0So taking small molecule<a href=\"http:\/\/www.greenray4ever.com\/lifexnotes3b2.html#TAALPHABETICSELECT\"> telomerase activators <\/a>effective at increasing the telomere length of components of the blood was a very good bet for effective life extension. Similarly, when dermal fibroblasts go senescent, they begin to attack the <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4e.html#EXTRACELLULARMATRIX\">extracellular matrix<\/a>, producing<a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4w.html#WRINKLES\"> wrinkles<\/a>. This does not happen at once to all of the fibroblasts, but gradually in a way that produces more and more defects in the extracellular matrix behind wrinkles. Thus a telomerase activator effective on dermal fibroblasts should prevent observable signs of old age such as wrinkles.<\/p>\n<p><span style=\"color: #000000\">\u00a0<\/span><\/p>\n<p dir=\"ltr\"><strong>When did you start your program? \u00a0What was your program in the beginning?<\/strong><\/p>\n<p dir=\"ltr\">I started taking telomerase activators on<a href=\"http:\/\/www.greenray4ever.com\/pic2007may.html#MAY2007B\"> May 1, 2007<\/a>, after reading material on <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4te.html#TA65\"><span style=\"color: #000000\">TA-65 <\/span><\/a>from TA Sciences. \u00a0I also had an exchange with Greta Blackburn of TA Sciences in which she emailed me promising results obtained by using<a href=\"http:\/\/www.greenray4ever.com\/lifexnotes3b2.html#81s\/6c\"><span style=\"color: #000000\"> TA-41<\/span><\/a>, an <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4an.html#ASTRAGALUSEXTRACT\"> astragalus extract<\/a> which they believed would be similar in effect to TA-65.<\/p>\n<p dir=\"ltr\">At first I expected to be able to set back my telomere clock by up to 8 or 9 years per year, but subsequently revised this to about 5.1 years per year.<\/p>\n<p dir=\"ltr\"><span style=\"color: #000000\">The <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4an.html#ASTRAGALUSEXTRACT\"><span style=\"color: #000000\">astragalus extract <\/span><\/a>actually finally turned out to work rather better, in fact, than<a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4te.html#TA65\"><span style=\"color: #000000\"> TA-6<\/span><\/a>5.<\/span><\/p>\n<p dir=\"ltr\">I was also reading <a href=\"http:\/\/www.lef.org\/\"><span style=\"color: #000000\">Life Extension Magazine <\/span><\/a>since 2004, so I was familiar with supplements recommended by LEF to prevent diseases of old age, such as<a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4am.html#ACETYLLCARNITINE\"><span style=\"color: #000000\"> acetyl L-carnitine<\/span><\/a> with<a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4am.html#ALPHALIPOICACID\"><span style=\"color: #000000\"> alpha lipoic aci<\/span><\/a>d for mitochondria, <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4an.html#ARGININE\"><span style=\"color: #000000\">L-arginine<\/span><\/a> as a <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4n.html#NITRICOXIDE\"><span style=\"color: #000000\">nitric oxide<\/span><\/a> booster for rejuvenating the vascular endothelium,<a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4an.html#ANTICANCER\"><span style=\"color: #000000\"> anticancer<\/span><\/a> supplements such as <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4g.html#GARLIC\"><span style=\"color: #000000\">garlic<\/span><\/a> and<a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4g.html#GINGER\"><span style=\"color: #000000\"> ginger<\/span><\/a> and<a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4tf.html#TURMERIC\"><span style=\"color: #000000\"> turmeric<\/span><\/a>, and so forth.<\/p>\n<p dir=\"ltr\">At first I took <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4an.html#ASTRAGALUSEXTRACT\">astragalus extracts <\/a>such a Solaray Astragalus Extract, and tried GAIA Herbs astragalus extracts. \u00a0Later, I got the idea of using<a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4cb.html#COLOSTRUM\"> colostrum <\/a>skin creams for telomerase activation. However I seem to have used astragalus extracts both orally and on my skin in the beginning. \u00a0This was done for two weeks before switching to a<a href=\"http:\/\/www.greenray4ever.com\/lifexnotes2.html#TELOMERASEACTIVATION\"> telomerase-inhibiting anticancer phase of treatment<\/a> for the next two weeks, when I took medicine like <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4cb.html#CURCUMIN\">curcumin<\/a>, which is anticancer. If my<a href=\"http:\/\/www.greenray4ever.com\/lifexnotes3b2.html#TAALPHABETICSELECT\"><span style=\"color: #000000\"> telomerase activators<\/span><\/a> tended at all to encourage <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4ca.html#CANCER\"><span style=\"color: #000000\">cancer<\/span><\/a>, I was going to squelch that every monthZin November 2009 I presented a short description of my program <a href=\"http:\/\/greenray4ever.com\/photonov1o2009.html\">here<\/a> For 2 1\/2 years, I had relied primarily on astragalus extract for telomerase activation. However, I also got 5 grams of arginine per day to improve my nitric oxide levels and improve the length of endothelial progenitor cell telomeres. \u00a0By June of 2009 I concluded after studies of Vida Institute literature that 33 grams\/day of astragalus root would serve as well or better than the 5 mg\/day of astragalosides from astragalus extract that I computed I was taking. So I switched from pure astragalus extract like TA-41, which worked fine according to Greta Blackburn at TA Sciences, to astragalus root in capsules from NOW FOODS. However, I was still applying astragalus extract directly to my scalp.<\/p>\n<p dir=\"ltr\">[Editor\u2019s note: Astragalus is a woody root which can be found in Chinese groceries for about $10\/lb. \u00a0Traditional use would be to boil strips of astraglus to make a tea. -JJM]<\/p>\n<p dir=\"ltr\"><strong> What is your supplements regimen at present? \u00a0Is it still evolving?<\/strong><\/p>\n<p dir=\"ltr\">I have posted a short summary of my present program and photo news <a href=\"http:\/\/greenray4ever.com\/pics2013may1.html\">here<\/a>. It is still evolving,\u00a0and by now I have 173 items at least on <a href=\"http:\/\/www.greenray4ever.com\/lifexnotes3b2.html#TAALPHABETICSELECT\">my list of telomerase activators<\/a> that I am exploring. The numbers of the activators chronicle the order in which I found them in the literature, or were tipped off to them by other investigators, except for sub-items, which I have sometimes found more frequently.<\/p>\n<p dir=\"ltr\">I have continued a four-week cycle featuring telomerase activators for two weeks followed by anticancer telomerase inhibitors for two weeks. \u00a0I do things that permit cells to recover from the senescent state that are a little unusual now: I use folic acid in my colostrum solution with an amphipathic alcohol to improve absorption (rubbing alcohol = isopropyl alcohol). [Isopropyl alcohol is moderately toxic. &#8211; JJM] \u00a0I take carrots and exercise to inhibit P16INK4A, which can stop senescent cell recovery.<\/p>\n<p dir=\"ltr\">I have many ideas about medicine I would like to include, such as <a href=\"http:\/\/greenray4ever.com\/photonov1o2009.html\">tocotrienol-rich fraction<\/a>. [These are chemical cousins of vitamin E derived from grains -JJM] \u00a0For bodybuilding, I take whey protein and creatine monohydrate, although I am not attempting a championship program. I can more easily resemble a gymnast.<\/p>\n<p dir=\"ltr\"><strong>Besides supplements, what are you doing to stay young?<\/strong><\/p>\n<p dir=\"ltr\"><a href=\"http:\/\/www.greenray4ever.com\/lifexnotes3b5.html#EXERCISE\">Exercise<\/a> improves circulation in the brain, elevates nitric oxide levels that dialate vessels, and provides numerous <a href=\"http:\/\/www.greenray4ever.com\/indexlongevity4ht.html#HUMANGROWTHFACTORS\">growth factor telomerase activators <\/a>that can be separately enhanced by various nutraceuticals. So I work out with weights, about 6 sets per muscle, using a split routine that takes 3 or 4 days. \u00a0This might be dangerous to a person with too much atherosclerotic plaque, or someone with glycation stiffening in the veinous system from high levels of sugar from ice cream. However, it tends to lower triglycerides and improve the lipid profile, and also to improve confidence and mood.<\/p>\n<p dir=\"ltr\"><span style=\"color: #000000\">I take a slice of Swiss cheese often to get a higher vitamin K2 level to avoid calcification of the arterial system and aortic stenosis.<\/span><\/p>\n<p dir=\"ltr\"><strong>In what ways do you feel younger than 7 years ago?<\/strong><\/p>\n<p dir=\"ltr\">May 2007 was my starting date, and we are now approaching May 2014. \u00a0I would say that my confidence has improved that we can finally master aging, although there is still a fair amount of suspense involved. \u00a0My physical condition oscillates somewhat, and is subject to minor perturbations. I work to get leaner in the Spring, like most people, and my self-image improves then. I look better in photographs, although I invariably puff up a bit in the Winter. In the Summer I can do pull-ups on outside chinning bars, and I get more V-shaped. \u00a0\u00a0\u00a0\u00a0In general, I seem to look somewhat better to myself in photographs an I did 7 years ago. Last Summer I did 130 pushups, which was \u00a0\u00a0\u00a0an all-time record for me. I am hoping that it will go back up to that this summer.<\/p>\n<p dir=\"ltr\"><strong>I notice you pay attention to appearance as well as health. \u00a0Do you think there&#8217;s a connection between anti-aging skin treatments and longevity? \u00a0Or a psychological effect whereby looking younger helps you to feel younger and programs your mind to live longer?<\/strong><\/p>\n<p dir=\"ltr\">I could dress better to improve my appearance, but my funds vanish into supplements. \u00a0Wrinkles are definitely symptomatic of the formation of senescent cells which begin to excrete factors that attack the extracellular matrix and weaken it. A weaker extracellular matrix leads to more than cosmetic problems, eventually, and less resistance available to UV radiation from the sun. \u00a0So keeping the skin healthy and young is very important for survival as well as for image. Skin cancer kills. Thin skin and failing veil cells around veins can produce bleeding, finally, perhaps even dangerous internal bleeding.<\/p>\n<p dir=\"ltr\">Of course, looking younger creates a sense of optimism about what may be possible, and helps reinforce a positive attitude.<\/p>\n<p dir=\"ltr\"><strong>What evidence do you have that your body is really younger?<\/strong><\/p>\n<p dir=\"ltr\">Unfortunately, I was unable to do measurements of my blood component telomere lengths because of funds could not be mobilized in time. Therefore, about all I have is my collection of <a href=\"http:\/\/www.greenray4ever.com\/agetransformation.html\">photographs online<\/a>. There are many measures of aging and senescence that may be applied to the problem of measuring the effect when funds are available. \u00a0See <a href=\"http:\/\/www.greenray4ever.com\/lifexmortality.html\">http:\/\/www.greenray4ever.com\/lifexmortality.html<\/a> .<\/p>\n<p dir=\"ltr\">&#8212;&#8212;&#8212;-<\/p>\n<p dir=\"ltr\">* I wish I could say, \u201cTesting experimental drugs on unsuspecting humans is off the table, out of the question, never done.\u201d \u00a0The truth is that it is not as exceptional as we might wish. \u00a0And the institutions that resort to surreptitious experimentation are usually up to no good.
