Defective Trigger for Muscle Growth, Survival Implicated in Heart Failure

Researchers have determined the molecular machinery that triggers normal cardiac muscle growth and survival, and have linked defects in this complex to an inherited form of human cardiomyopathy, a type of heart failure where an enlarged heart loses its ability to pump blood. Published in the December 27, 2002 issue of the journal Cell, the study also identifies a subset of German cardiomyopathy patients with a specific gene mutation that disrupts the heart muscle’s normal stretch activity. This mutation has apparently been passed down through several generations of Northern Europeans.

Cocaine harms brain’s ‘pleasure center’

New research results strongly suggest that cocaine bites the hand that feeds it, in essence, by harming or even killing the very brain cells that trigger the “high” that cocaine users feel. This most comprehensive description yet of cocaine-induced damage to key cells in the human brain’s dopamine “pleasure center” may help explain many aspects of cocaine addiction, and perhaps aid the development of anti-addiction drugs. It also could help scientists understand other disorders involving the same brain cells, including depression.

New study ties moderate beer drinking to lower heart attack risk

A beer a day may help keep heart attacks away, according to a group of Israeli researchers. In preliminary clinical studies of a group of men with coronary artery disease, the researchers showed that drinking one beer (12 ounces) a day for a month produced changes in blood chemistry that are associated with a reduced risk of heart attack.
Their study adds to growing evidence that moderate alcohol consumption may reduce the risk of heart disease, the number one killer in the United States.

Possible New Treatment Strategy for Deadly Brain Tumors

Despite advances in neurosurgery and radiation techniques, the prognosis for patients with intracranial glioma remains devastating. Now, researchers have identified a possible new treatment strategy for this common type of malignant brain tumor. Two studies funded in part by the National Institute of Neurological Disorders and Stroke (NINDS) show that, in a mouse model, neural stem cells (NSCs) can be used to deliver therapeutic agents capable of killing glioma cells and their migrating tumor cells.

Gene mutations in breast tissue may make cancer detection more difficult

Until now, researchers thought that breast cancer nearly always began when cells in the epithelium went haywire. But new research suggests that genetic mutations can ? and do ? occur initially in a deeper layer of breast tissue, called the stroma. This presents a serious concern for physicians, who believed that breast tumors spread from epithelial tissue. “Genetic alterations in carcinomas, including breast cancers, have always been attributed to epithelial cells,” said Charis Eng, Klotz professor and director of the Clinical Cancer Genetics Program at Ohio State University. She co-authored a new study that looks at genetic mutations in breast tissue.

Scientists have long recognized that, despite physical differences, all human populations are genetically similar to one another. But a new study in the journal Science concludes that populations from different parts of the world share even more genetic similarities than had previously been assumed. At the same time, researchers found that tiny differences in DNA can provide enough information to identify the geographic ancestry of individual men and women.

First Virtual Stomach Explores Drug Delivery

Researchers have developed the first virtual stomach, a computer-generated model that is providing unique insights into the way medicines are released from pills and capsules. “There’s no other technique to allow you to see flow patterns in a real stomach, animal or human,” said lead researcher James G. Brasseur, Ph.D., a professor of mechanical engineering and bioengineering at The Pennsylvania State University. Although pills and tablets have become a ubiquitous part of our society, the detail of how the stomach breaks them down to help release their medicine remains unclear. “You can give someone a drug and measure its uptake in tissue,” he said, “but the process between administering the drug and the uptake is largely guesswork.”

Scientists Develop New Gene Therapy Approach

Researchers have developed a new gene therapy approach that prevents the AIDS virus from entering human cells. The technique offers a potential way to treat HIV patients and could apply to any disease caused by a gene malfunction, including cancer. The research team created a new application for a genetic technology called small interfering RNA (siRNA). The synthetically designed siRNAs act as a catalyst to reduce the expression of specific genes and slow the progression of disease.

Gene Responsible For Anemia (Type CDA-1) Discovered

A combined effort between Israeli scientists has led to the discovery of a gene responsible for a type of anemia primarily found in a number of Bedouin families, called congenital dyserythropoietic anemia-1 (CDA-1). The findings, published the December issue of The American Journal for Human Genetics, could lead to effective detection and eventually treatment of the disease. In addition, understanding the role of this gene?s protein product in the body could provide important clues to other types of anemia, as well as to the general mechanisms of blood cell formation.

Transplanted tissue could offer a solution to kidney donor shortage

Instead of searching for a kidney donor, a new study suggests, one might be able to grow a new kidney. A team headed by Prof. Yair Reisner of the Weizmann Institute of Science has induced human stem cell tissue to grow into functional kidneys, and have accomplished the same with porcine stem cell tissue. Published in Nature Medicine, the method could lead to a promising solution to the severe shortage of kidney donors.

Designing a robot that can sense human emotion

Forget the robot child in the movie ?AI.? Vanderbilt researchers Nilanjan Sarkar and Craig Smith have a less romantic but more practical idea in mind. ?We are not trying to give a robot emotions. We are trying to make robots that are sensitive to our emotions,? says Smith, associate professor of psychology and human development.
Their vision, which is to create a kind of robot Friday, a personal assistant who can accurately sense the moods of its human bosses and respond appropriately, is described in the article, ?Online Stress Detection using Psychophysiological Signals for Implicit Human-Robot Cooperation.? The article, which appears in the Dec. issue of the journal Robotica, also reports the initial steps that they have taken to make their vision a reality.

New approach holds promise for reducing cocaine craving

New understanding of the changes in brain chemistry caused by chronic cocaine use has suggested a novel treatment that could reduce the intense craving that forms the core of cocaine addiction. “Our studies show that administration of an existing drug ? n-acetyl cysteine, which is used to treat cystic fibrosis and several other disorders ? reverses the changes in brain chemistry that appear to cause cocaine craving,” said David A. Baker, a post-doctoral fellow in the laboratory of Peter W. Kalivas at the Medical University of South Carolina.
